You should check this:

Make sure battery is charged.
Connect USB cable to USB port which is directly to motherboard.
Try other USB cable.
Drivers may not be fully installed.
Drivers may not be fully loaded.
Other computer might solve this problem.
Other OS may solve this problem.

If you are using Windows 7 64bit, you should try to run DC-Unlocker as administrator (right click>Run as administrator).
